Rafael Marquez is probably the most talented player on the Mexican national team. I actually think he plays better for Barcelona than he does for Mexico given his history of red cards (2002 World Cup, 2005 Confederations Cup) but he's talent nonetheless.
So it will be difficult to replace him for the Gold Cup if he gets his way and stays with Barca for the duration of the year.
Mexico has no one that can replace him. Carlos Salcido and Ricardo Osorio will have to step up their game and take some of that leadership role over. Joaquin Beltran, a Hugo Sanchez favorite, might step in and take over for Marquez but that's not nearly enough cover. It'll be interesting to see who gets that role should Marquez stay with Barca.
